I think all Doc Ock needed to overcome his career of crime was a good woman. If he had someone to focus his attention on for valentines day and to tell him that his master evil plans were silly and he needed to fall in line, he would have retired from the evil master mind thing years ago. If I were Spider-man and I wanted to finally end his evil career, I would have worked with him on setting up dates.
I don't think it would happen, realistically, if Peter and MJ are happily married. In order to do this, Slott had to have a status quo where both Aunt May and MJ are pretty much out of the picture of Peter's day to day life and he has no current girlfriend. Peter had to be isolated and alone for it to work. And even with those elements in place, he still has Carlie and MJ suspecting. I don't think Aunt May has met SSM yet.
Theoretically, yes you could do it. Like Quesada's argument about the married years being the same with an unmarried Peter and MJ. Yes, theoretically its possible but would it happen the same if it really were that way? Hell no. The purpose of the whole 1990s Clone Saga was to bring back a single Spider-man. Without the marriage in place, the 1990s Clone Saga doesn't even happen probably. Same with dozens of other stories. Just because writers could do something does not mean they would do something.
So no, if Peter and MJ were still married I don't think we would have had SSM. Is that really a bad thing? Slott or another writer would have come up with something else equally cool.

Perfect humility dispenses with modesty.
If they were married, the whole "rape" crew would have gone bananas since it's inevitable a married couple would do some romping.
Crazy fans would scream raaaaaaaaaaaaaaaaaaaaaaaaaaaaaappppppppppppppppeee eeeeeeeeeee even more than they did before.
It took Spock beating the holy hell out of jester and screwball to make Mj realise that it wasn't Peter even before with the drinking and the calling her woman she was oblivious which is annoying given she realised past fakes straight away.
Being married would make MJ being oblivious even more frustrating almost as frustrating S when peters memories drew an octopus and black widow thought it was a bad picture of a spider.
Married? MJ beats Spock with a bat til he decides she is Superior and gets the hell outta there
At first I was thinking you needed it, but given Mary Jane's chosen profession, you could still probably get away with it. It's certainly the strongest candidate for needing OMD (five years later....), but it's also true that since she's a model/actress it's fairly easy to get her out of the country/city/book for a few months or a year of Marvel time. With that said, you have to get rid of her fast uness you want to deal with the consequences/implications of them living together, and it'd come off as awfully convenient.
Though needing OMD to have Superior is like needing an open wound so you can get an infection, but eh.

What Would Spidey Do?
No difference. People would have still jumped to the "rape" conclusion, only this time, they wouldn't have needed Slott telling them that MJ would be with the "new Spider-Man" plus that SSM #2 cover with Spock forcibly kissing a stunned MJ, something that thankfully didn't even happen in the actual issue.
