    Off the top of my head...

    I'd add (Peter Parker:) Spider-Man #75 -- It reads well, and it's important in that it sets up so many things that are a part of the "modern" Spider-Man. And it has a lot of the answers to past questions.

    I'd also add Ultimate Spider-Man #13 -- where Peter reveals to MJ that he's Spider-Man. So cool; I reread it at least once a year.

    I would just include the entire 'Learning Curve' TPB, which covers issues 8-13, including the brilliant confrontation with Kingpin and ending with the reveal to MJ. It captures the spirit of USM (and a modern take on Spider-Man).

    Amazing Spider-Man 226-227 - This two-issue story captures the essence of the modern Black Cat character. Stern's take on her really stuck around.
    Perfect humility dispenses with modesty.

    Some that haven't been mentioned yet:

    Amazing Spider-Man #17-19: (Aunt May sick, Spidey flees from Green Goblin and is thought a coward)

    This pretty much has everything. Green Goblin really comes into his own in his second appearance. The first real multi-part saga in the series, a lot of recurring themes of the series started here. Really groundbreaking stuff.

    Amazing Spider-Man #25: Captured By J. Jonah Jameson!

    The first full on comedy issue, and still one of the best. A flawlessly told farce.

    Webspinners: Tales of Spider-Man #7-9: The Bridge (Peter's high school prom)

    Peter gets tired of being pushed around and decides to put himself first. And gets three prom dates. And it all goes horribly wrong. Gets to the core of teenage growing pains and responsibility themes without getting too dark or glum. And there are some good laughs along the way.

    Amazing Spider-Man #583: Platonic (Betty Brant/Peter Parker friend times issue)

    Peter Parker through the eyes of a friend who doesn't know that he's also Spider-Man. A very good character study of Peter's flaws and strengths.

    Amazing Spider-Man #665: Crossroads (Betty Brant hospitalised)

    Another short story all about guilt, decisions and a conflict of responsibilities. Very poignantly shines new light on an aspect of Amazing Fantasy #15 without shoe-horning or retconning anything in.

    Untold Tales of Spider-Man '96 A Night on the Town (Spider-Man goes on a date with Invisible Girl)

    It's fun, it's funny, and it's not often we get to see Spider-Man drawn by Mike Allred.

    No One Dies, ASM 655-656, is a modern day classic.

    Spider-Island, ASM 667-673 is the best Spider-man event story, jammed with character moments and action in a solid, fast paced story.

    And just about anything DeMatteis did in Spectacular. The highlight was probably the Green Goblin stuff or the Vulture 3 parter.

    Amazing Fantasy #15

    Amazing Spider-Man #1-89 (deliberately leaving it there, after all, Stacy's deaths, George and Gwendolyn, is overrated, and just plain death...YEEEAAGHHH!!...but still enough for some intense Doctor Octopus action)

    Amazing Spider-Man Annual #21 (The Peter Parker cover, with all his friends, in he and MJ's wedding, as it should be)

    That shall be enough, for the basics.

    My personal favourites. I think anyone reading the character for the first time, would get a pretty good idea what he's about after reading these 100 stories.

    Spectacular Spider-Man from the late 80's was pretty good. It was really dark for Spidey stuff!! Tombstone and bulimia are what stick out to me most.

    I add on some of JMS' run. His early issues were really solid. As was his reunion with MJ. The talk between Aunt May should be on the list. I'm no good at issue numbers though.

    I'm really not a random issue reader though. I enjoy a good run of comics.
