which is better or more collectible?
It all depends on the character. "Better" is subjective and people will have different answers. "More Collectible" would be determined by market rates and what a buyer is willing to pay, which again, will vary by character.
Appearance.
Take Wolverine. He appears in Hulk 181 (or whatever), then in Giant Sized X-Men and All-New and All-Different X-Men. His first solo mini wasn't until 1982 (long after he was established) and then Marvel Comic Presents and Wolverine came out in the late 1980's for his solo.
Hawkeye, X-Men Legacy, FF, New Avengers, Uncanny Avengers, Animal Man, Wonder Woman, Manhattan Projects, Fatale, East of West, Bedlam, Saga, Bandette
to clarify i guess i am talking more golden age stuff for 2nd tier characters so to speak
for example: son of satan in which his 1st app is Ghost Rider #2, he was in Marvel Spootlight #12, and his own series Son of Satan - all in the bronze age
Spider-Woman: Marvel Spotlight #32 1st app then own series Spider-Woman - all in bronze age
I would like to buy a 1st issue from these characters but don't feel i need to get more than one issue
As for the above mentioned Wolverine, i guess this character is too major to "either or" for 1st app or 1st issue. I have 181 and #1 from his '82 mini solo. Just like Silver Surfer. I want his silver age #1 but also have his #1 mini from early 80's
I'd go for both to be honest. The first appearance is rare for me, its like a glimpse of Thanos at the end of avengers. Its a little thing which leads to something massive.

The first appearance is always more sought after.

Which is why I still have some hope of completing my Captain America and Iron Man runs. Since the first issues of each of their original series aren't their first appearances, the prices have not gotten so astronomically high as to keep them out of my reach.
However, the one that irritates me to no end is Moon Knight. C-List character at best, but dang near impossible finding his first appearance in high grade that you won't have to give up serious $$ for.

Currently Hunting: Captain America # 117 (last one for the entire run)
there's no all encompassing rule that can be applied to all comics to determine how valuable they are. If you know what issue you want, find out what people are asking for it, and find out what collectors are paying for it.
Still, for the most part, the most valuable issue a character is in is the one with their first appearance.
