Spidey would be nicer and actively try to change things for the better. However, if the What if? where he retains Captain Universe's power is any indication it might also strip him of his humanity.

Honestly, I think Pete would crack under the pressure. Not to mention the whole 'cosmic balance' stuff that would probably require Pete to make decisions that run against his grain.
Omniscience is such a character defining trait, I expect that he will ultimately be pretty much the same.

Hmmm... I'm not sure if this will cause Spider-Man to suffer more or less. I would have said the need to make hard decisions going against his basic morality could cause him to suffer... but (and I don't like to refer to this off-hand), he did make a deal with the Devil. Maybe eventually he'd break down and embrace it.

Obviously, the first thing that will happen is that he will run into a universe of Uncle Bens, Aunt Mays and Mary Janes that is threatening to accidentally and unavoidably unbalance the cosmos.

Well, biggest question first: Are we talking current spider-Man? Because if we are, we're all screwed.
If we're talking classic Spider-Man (i.e. - pre-3-weeks-ago), well, the biggest issue is that Pete is going to do a Beyonder and off Death first thing. Pete's biggest thing is "nobody dies".
No, it's not. It's a big thing, no doubt. But his biggest thing is "With great power comes great responsibility."
If we extrapolate that, then: With nigh-absolute power comes nigh-absolute responsibility.
Pete's responsibility would be to maintain the way things are supposed to be in the Universe. He'd probably accept that, regretfully.
It would hurt his brain A LOT, but I'm assuming that with this power would come a degree of mechanism for dealing with (or completely losing) that moral stance and associated mental anguish. Otherwise it would be a really dumb move by TOAA to give the job to anybody at all.

I see him becoming like Death of the Endless: Compassionate, knowing, but ultimately acting with a higher understanding of universal purpose.
At that scale, would Death even matter to him? I mean, its gone from "people being lost for the rest of their loved ones' lives" to "people going into another room slightly earlier than their loved ones inevitability go into that same room."
He could probably spend any free time having tea parties with Uncle Ben, his parents, the Stacys and his marriage.
