Well, Gennosuke's dojutsu is going to make it hard for Manji to kill him since he has no notable defense against hypnosis.

However, Gennosuke is not going to be able to kill Manji because of his immortality. He can likely incapacitate him though. Have Manji chop himself to pieces and then bury him or something.
