I think characters THAT obscure might be a bit of a push (though Plastic Man and Hawkgirl are a far-out possibility), but I do get the impression that they're rolling out the big guns now and saving more obscure character reveals for later (I'm thinking Animal Man and Aquaman are sure things for this). From an advertising perspective, you want to bring in the well-known faces and the ones getting the push (Cyborg, Green Arrow, etc.), THEN bring in the underdogs/obscure fan characters.

That said, if Etrigan the Demon or Sir Ystin are in this it would make my life, haha.

Considering the amount of freedom all the NetherRealm guys are claiming they've gotten from DC, I imagine they'll include characters they want based on the personal desires of their team, as opposed to being too concerned with how obscure some characters might be. They had DLC for Freddy Kruger in the most recent 'Mortal Kombat', well after his cultural relevance had peaked. Hell, I would have never guessed Solomon Grundy would be included in the game, let alone been a character included alongside Batman and Superman in the initial reveal.

My money would be on Brainiac as the final boss, though I'm kinda surprised that Luthor hasn't been mentioned yet.

How could I have forgotten J'onn! He'd be awesome to have in the game. I think CC has been strongly hinted at, an Mirror Master with teleporting and/or duplicate reflection moves'd be awesome too.

I know Batwing has been hinted. Nothing against Batwing, I enjoy the character but there are already enough characters from the Bat family. If they wanted to make the line up more diverse there are so many great characters to use. Really hoping for Blue Beetle. His powers would be so much fun to use in a fighting game. Mr. Terrific would be great as well. I would love to KO an opponent and then have Michael say "fair play". :)

I think Aquaman and a Green Lantern (I'm going to assume it will be Hal with at least one alternate skin being either John, Kyle or Guy) are sure things. With the fighting system that has been shown thus far (and how much I still play MK9), I'd love to see Hawkgirl, Martian Manhunter, Plastic Man, Black Canary, Animal Man, Blue Bettle, Booster Gold (who I think probably has a higher chance of appearing if they want a Johnny Cage-esque fighter) or Swamp Thing added in. Villian wise: Mirror Master, Captain Cold/Mr Freeze, Deadshot or Toyman.

Not necessarily. Many of the alternate skins in Ultimate Marvel vs Capcom 3 were different characters (examples Red Hulk for Hulk, Red She-Hulk & Savage She-Hulk for She Hulk, Iron Patriot for Iron Man, Bucky's version of Captain America for Captain America off the top of my head) but they still had the same voices.
