Transmetropolitan vs. Preacher?
Disregard the "vertigo vs. preacher" question if you will.
Preacher - but they're both great.
I just read all the Preacher volumes, and only read Transmetropolitian vol 1 so far, so I can not say which series is better, but Preacher started off very strong, and dipped in quality in the middle of the run, but the last volumes were better, but I found the ending very anti-climatic. But overall I enjoyed reading it, and would recommend it.
I'm pretty new to comics too, and for me it's interesting to hear and read about what people think are classic, great comic books - so that I can use these as a reference when I start to read other, newer stories. Because of this I picked up a couple of volumes of Sandman to read it from the beginning, and to be honest, it wasn't like something jumps out of the page to punch you in the face and tell you that it was the most awesome thing you've read, ever. I think the reason why people say that these stories are classic is because they give you plenty of food for thought. You read the book, then you put it down; and then you reflect on it and that's when the good points of the story stick with you.
That being said, the first six volumes of Sandman have some parts that are connected to a bigger story, and much of the writing may be past what I can understand. At times though, you jump from what seems like one genre to another. But it's a story that will make you think.
Changing the subject, the other story that I've been following is Hellblazer. The most fun thing about this story is seeing how much of a mess Constantine is, as well as seeing the interpretations that writers take on demon lore.
Anyways, I guess what I was trying to write here is that these "Must Read" lists are a lot like the tier lists that you find for different fighting games. It doesn't mean that just because everyone says these books are the best that they will be the ones you enjoy the most.

Also, Batman & The Joker showing up in the first actual arc of the series is pretty much half due to stirring up sales/interest & half 'It's bloody Gotham City; if we gotta do this, let's just get it over with soon as possible.' But even then, I'd say Hitman Book 1 is the weakest mostly by default.

I loved this series and looked forward to it as it ran. The story is really good and the art is outstanding. It's one of those titles with individual panels that remind a reader why they got hooked on comics in the first place.
I have all the individual books, but I'll probably grab the trade when it comes out just to have it.

Thirding or fourding or whatever the Hitman recommendation. First trade is indeed the weakest (Demon-annual and terrible crossover tie-in, Batman-terrible crossover tie-in...), once the series gets down to Tommy/Natt, Tommy/Tiegel, Tommy/Ringo and so on, it gets good.
