Why aren't you reading Winter Soldier? You should be!
I was thinking the same thing. Also, it seems that this film may be taking a cue from Ang Lee's Hulk movie in that it's strongly suggesting that Peter's dad may have been the one who created the spider that gave Peter his powers. Which I REALLY hoped they weren't going to do with this whole "mystery" regarding Peter's parents.

POSSIBLE SPOILERS FOR THE AMAZING SPIDER-MAN BELOW
Three sources have told me this, but I held it. I didn't see the point in running it now, when the movie has been shot and edited and is ready to go. Nothing can be changed. But then the new trailer hits today and all but spells this out, so what the hell, here we go:
Spider-Man's origin has been changed in The Amazing Spider-Man. The spider bite no longer gives Peter powers, it rather activates something already in his genetic structure. Something placed there by his father. Peter Parker is no longer super-powered by chance, but rather by design.

'nerd, we finally agree on something. The "origin" not being an accident is beyond bogus. So, so disappointing.

HaHahahahahahahahahahahh!!! but you got'cher mechanical webshooters!
My guess is that it's Irrfan Khan's character. He was originally cast as Van Adder, AKA Protogoblin, although the official website says that he's playing Dr. Ratha. So it seems to be someone mysterious with ties to Osborn.
It makes sense to use either an obscure or new character. It allows Sony to have some foreshadowing, without yet having to cast one of the biggest roles in the sequel.
I'm guessing Oscorp is trying to build the first superman pulled from the Ultimate U origin.
