I was thinking of how Kraven's Last Hunt was for Kraven and then thought of Electro... Was Light the Night Electro's defining storyline? How about the rest of Spidey's rogues?

The difference is Light the Night told more about Eletro and his life where Kraven Last Hunt still was focus on Spidey's life kinda. There wasn't the back drop on Kraven life Like there was on Eletro. That was my take on the two storylines.

This is difficult as you have to choose great stories that have defined how later creators interpret them. Sometimes it's the first appearance/ sometimes it's not.
Green Goblin 1- Marvel Knights Spider-Man #1-12. Note- It's not as good as "The Death of Gwen Stacy" but it gives a better sense of Osborn/ The Green Goblin as characters.
Harry Osborn- Spectacular Spider-Man #178-200
Venom- Amazing Spider-Man #300/ 315-317
Doctor Octopus- Fantastic Four #267 "A Small Loss" It was during the period when he was afraid of Spider-Man, and reveals his brilliance/ insanity/
Kingpin- Daredevil #170-172. When he becomes Daredevil's arch-enemy.
Daredevil #227-233: Best Marvel comic ever.
Hobgoblin- Amazing Spider-Man #238-239. Established him as a brilliant guy willing to learn from osborn's mistakes.

A few more.
The Lizard- Amazing Spider-Man #101-102. Probably the best early example of the Lizard as a great enemy of Spider-Man/ Connors as a great ally.
The Vulture- Amazing Spider-Man #240-241. He finally gets an origin, and is defined forever as a vicious powerful old man.
Mysterio- Amazing Spider-Man #13/ 24. Perfect portrayal of a brilliant man willing to drive Spider-Man insane/ confuse his enemy at all times. Just like the good Mysterio stories told later.

I think, for the villain duo of Leather and Lace, probably their defining moment happened in a story that Karl Kesel and Patrick Zircher did in Web of Spider-Man Super Special #1.
I can honestly say that I never liked any Leather and Lace story any better than I liked that one.

Also, dirung the Mysterio story in asm #196-199, the Kingpin finally beat Spiderman in asm#197, defining him as a great villian ready to move to challenge somebody else (daredevil).

Doctor Octopus' one would have to be Spider-Man Unlimited #3 or the Web of Death arc during the Clone Saga.
Both are great Ock stories. And very deep ones which show a side to Octavius that we never saw before.

Add Stern's HOBGOBLIN LIVES as a bookend to his origin & first appearances in ASM, as HL finally reveals who Hobby really was, fleshes out many of his behind-the-scenes machinations, and cleans up all the Leeds/Macendale nonsense that had dragged his good name through the mud.
