The two mainstream (I know 616 is the mainstream universe but Ultimate and 616 are the two most popular) universes have two totally different Spider-Men. Besides race what do you think their differences are? What about their costumes? Their reasons for being Spider-Man? Their personality? Their powers? So, discuss!

I guess we can compare origin stories, but, yeah... not much point on anything else, though to Miles' favor, he did knock out Ultimate Electro on his second fight. An honor last received by the ULTIMATE AUNT MAY (Should-Be Director of S.H.I.E.L.D.)

That summarizes popularity difference. Not so much the differences asked in the original post.

Miles seems to be more introverted than Peter was. That may change as he gets more experience, though. It would be better to ask this question in a couple years when Miles has a decent amount of Spidey experience under his belt.

Peter Parker IS the real Spiderman and the best character that Marvel has.

Miles is a new character, true, but what kind of a man is he? Has he made any "Devil Deals", yet?
Peter Parker IS the real Spiderman and the best character that Marvel has.

Umm, I think it is a little too soon to really compare the characters, as a previous poster has stated. I can't help but wonder if you're just very very sneakily trying to invite some Miles hate onto the Spidey board, since just about everyone on the Ultimate board seems to be onto your methods already. I mean, c'mon, did you honestly suggest that we discuss and compare their costumes? Just look at the pictures you posted. Everything that could be said is obvious, except for maybe an opinion such as "Miles' costume looks cool (which I do think)".
Still, I'll play. I haven't picked up issue 5 yet due to being out of town for the holidays, but I think Miles is very interesting. He was initially more reluctant about his powers, and feared being hated. He has a best friend that he immediately told, and he is living in a dorm. Honestly, we have a new Spider-Man, or a new character that has the same cool powers (mostly) as Spider-Man, and I find that exciting. I want to see how things develop with his supporting cast, and I want to see the mostly unique situation of Peter's loved ones finding out about the new Spidey and voicing their thoughts on the matter. I don't want Bendis to dwell on this, but I do find it very interesting, and after each issue he's got me hungry for more. I feel similarly about Kaine in his new series, but there we know even less right now.
616 Peter, the real Spider-Man (and I only say that because it's the same character, in continuity, that Lee and Ditko introduced almost 50 years ago) is like an old friend, a rock, a comfortable, welcome read. I like what Slott has been doing lately, and I think Pete has had a great year. Spider-Island was, to me, a shining example of how an event should go. Of course, I don't need one every year, or even every two years. I'd much rather have more good ol' Spidey stories, status-quo type stuff for a while. I enjoy his established supporting cast, his dishing out of hard knocks and funny (or eye-roll-worthy) quips, and the Parker luck.
Contrasted with Miles, however, is that I don't find Pete quite as exciting or interesting anymore because of the fact that Marvel clearly won't allow him to grow up anymore. I enjoy Amazing, but it's clearly the "safe" read, or it usually is. It's just that Pete has been forced into such stupid crap in the last few years, or last decade, or last two decades, all in the name of regression and for the (not-unanimous) desire for a swingin single Spidey. There have been great stories in these times as well, but I don't feel that they justify all that nonsense.
Look at Spider-Island. There was a great moment, a very sort of tragic, dramatic, angsty, shipper-type moment at the climax of the story, where it is made abundandtly clear that Pete and MJ belong together. And this after the marriage is so clumsily and stupidly destroyed. Thanks to the efforts of the powers-that-be at Marvel, we seem doomed to have a Spider-Man who can only engage in shallow, pointless (outside of maybe some non-romantic benefits) relationships in which he is never truly happy. Unless he gets back with MJ, of course. But then, that would never be allowed to last. So they'll be on-again, off-again forever, which is just ridiculous. They should have just killed her off, or gotten them divorced in such a way as that it is clear they should not be together, because to torture and tease the readers like this could be fun at the time, but in the big overall picture, it seems just plain stupid.
Hmm, big rant, I got more, but we'll let that one end right there.
I do enjoy Avenging Spider-Man so far as well. It's got great art (for now), and a large amount of plain-old pure Spidey fun, minus the Parker side of things (so far at least). The only downside is the price.
In summary, Miles and his all-new storyline is exciting and interesting, and I want more, faster. Peter is still a fun read, and you just can't beat that good ol' Spidey action, heroics, and quips, not to mention the Parker luck. But Pete's private life is in a bit of a shambles socially, and although this has always been somewhat the case, the reasons behind it this time (deal with the devil) are monumentally stupid, whilst before, it was common sense that his personal life would be difficult to juggle with the heroics. I think he should have been allowed to grow up a bit more.

I dropped Miles' book after issue four, because I found it rather boring. More to the point, I found Miles rather boring. Just about every character around him is more interesting than he is. His father, bestie, and uncle each have much more personality than poor Miles. He only reacts to situations, and didn't seem like he was going to take initative any time soon.
And Bendis' attempt to try and give him his own "Uncle Ben" moment, with Peter as Ben, while clever, doesn't exactly work. Yes, you had spider powers, and maybe, possibly could have done something to help save Peter. Or, you could have just screwed it up even worse. I'm six four and two-fifty (and dropping, yay), but if I pass a burning building, I'm not going to rush in after the fire fighters. Yeah, I'm physically capable of pulling a person out, but I'm much, much more likely to get myself killed. Guilt doesn't factor in to overwhelm common sense.
I think the opening character moments of both Ultimate Peter and Miles kind of set their differences. We first see Peter in a crowded mall, totally by himself, getting harassed by others. We see what makes him stand apart. We first see Miles in a crowd with hundreds of other students with a vested interest in the same process. We see what makes him blend into the background.
