What situation should happen in Spider-Man that has never been done before?
What changes could be made in Spider-Man that have never been done before?
I think they should do a year long arc where he's stuck in either a country or a planet. I dont think that's ever been done with him before.

There are a few things I'd like to see, but I'm hoping that I get to write the Spider-Man comics one day, so I wouldn't want to mention it now.
That said, it'd be kinda cool to see Spider-Man (the quintessential New Yorker) back in time in an earlier era of the city's history for an arc.
Marvel really hasn't explored Peter's family much. There's some potential there. Plus, Peter's parents being revealed as traitors after their death is a reasonable explanation for why his mother's side might have avoided contact with him.

Let's see a storyline where Jameson has Spider-man's powers and Peter is running the Bugle, even if it's just a illusion Mysterio set up.
Due to some time and space bending, Peter Parker finds he's got two new housemates that he's going to have to try to make fit in for an extended period of time. Those two would be the Ultimat version of himself (or a reasonable factsimilie) and Mayday.
Spider-man loses his powers for a while (not new, I know) and tries to rely on various gadgets and tricks he's come up with to fight crime.
Time travel story where Peter actually works with his parents on a mission al while trying to hide his identity from them.

Just as a short-term storyline, I wouldn't mind a "Spider-Man: Agent of SHIELD" story. Also, though I have no idea how they'd pull it off, I think a "Donnie Brasco" type of thing with Peter or Spider-Man undercover as a criminal would be cool.

Peter gets a job bar-backing at the Bar With No Name. One of the villains then hires him as their sidekick for some upcoming 'jobs'. Peter gets a first-hand look at the criminal underworld, and just how pathetic -- and in some cases, noble -- some of the villains are.

I think i'd introduce a super powered stalker that wants to be Spider-Man's side-kick, and keeps showing up halfway through a fight with a super villain, and always makes the fight go badly.
That or i'd make Peter homeless.
Or both!

i think some people would prefer him being homeless as opposed to living with aunt may ;)

At the moment he's doing neither. He's sharing an apartment with Vin Gonzalez now.

I mean really homeless. But too proud to ask Aunt May for help...

I'd like to see him done in a not quite Spider-Man way. Peter Parker as a cool, witty guy who works a decent photographer job, but wouldn't want anything to do with Spider-Man. Because Spider-Man is a freak. An awkward, mysterious, and kind of frightening Spider-Man, emphasizing his ability to combine logic, powers, and gadgets, focusing on crime, yet still mixing it up with the most bizarre super villains, maybe even out-creeping them. He'd still be a mostly daytime super hero and there wouldn't be any identity issues (I don't think that will ever work for Spidey), but we'd see all sorts of sides of the same coins.
....Or just dating an older woman.

Its actually pretty difficult to think of things Spidey hasnt done before....has he ever been on the run with one superhero riding shotgun..I'm not counting Civil War.....

I'd like to see brand new villains, brand new supporting characters. We have some but keep going, don't just continue with the ones just introduced, keep introducing new ones, revamping old ones.

One brand new thing I'd like to see...
Writers reusing villains introduced since the early 90s, especially those created by other writers. I wouldn't mind seeing more new villains, and it seems like we'll get some in the months to come, but I'd also love to see villains since Carnage with recurring roles, not just the Brand New Day guys.
Fusion. Digger. Carlyle Calimari. Shathra (or her children.) Morlun's assistant Dex. Even Charlie Wiedermann. I'm sure all these new guys have decent stories in them.

I always liked Coldheart. Stunner also really needs to make a come back.

I'd really like to see a story where Spidey goes to space and take on Doc Ock at a space station hideout he built. That's never been done before.

I think that happened when Superman and Spider-Man met for the first time.

I think that happened when Superman and Spider-Man met for the first time.It did. Spider-Man fought Ock and Luthor on the station while Superman took off to stop a tsunami from wiping out the east coast. Good story.

Here's a story I'd like to see, but probably won't. More of a sub-plot, really.
I'd like to see Peter starting to have a series of "dreams" where he is trying to talk to Uncle Ben, but Ben won't do it because he is ashamed of Peter. Peter will be trying to figure out why, but the audience will know it's Ben's disappointment with Peter's actions during OMD.

Since they wouldn't allow a Crossover between the Ultimate Universe and the regular Marvel Universe and since he meet the Spider-man of 2099, then why not have her meet Mayday Parker, a.k.a Spider-girl, his alternate future daughter.
Either that or unmasking him all over again to the people around him.
