I'm pretty sure Spidey got owned by the Vulture first when the Vulture dumped him in a water tower leaving him to drown.Correct me if I'm wrong.

You're not. Doc Ock is the first villain he loses to and doesn't defeat in one issue. My bad. The Vulture encounter is in Amazing Spider-Man #2.

I have two guesses, but I believe it's John Jonah Jameson.

Edit: The ones you missed, if they can be counted, were Hybrid, the combined symbiote of Phage, Riot, Lasher, and Agony; and She-Venom II, which was made using a cloned Venom symbiote.

That would get complicated, considering the Sinister Seven that came once and the Sinister Twelve Millar introduced. The original Six were Vulture, Mysterio, Doc Ock, Electro, Kraven, and Sandman. After Kraven died, I know he was replaced by Hobgoblin. During Mackie's run, Venom was part of the team. In Civil War, a Sinister Six was apparently gathered that had Doc Ock, Lizard, and, I think, Shocker. The other three weren't seen.

As for the Sinister Syndicate, Boomerang, Rhino, Beetle, Hydro-Man, and Speed Demon were the original members.

One other that I can think of is the costume shop version of the red and blues that Peter wore with a cellophane mask that he complained about after something happened to his original.( and just in time for Halloween)

As I recall they did something like this again in the storyline (mentioned in a Peter thought balloon) when Todd McFarlane took over the book to justify the radical difference in Spidey's appearance, they stated intially that it was a Halloween Costume.
